Combinatorial diversity of V H region was calculated to be ∼6,000. Of the 44 V H segments with an open reading frame, 39 are expressed as heavy chain proteins and 1 as mRNA, while the remaining 4 are not found in immunoglobulin cDNAs. The region contains 123 V H segments classifiable into seven different families, of which 79 are pseudogenes. They know there's another strand, and they know how to figure out what its sequence is if they need to.The complete nucleotide sequence of the 957-kb DNA of the human immunoglobulin heavy chain variable (V H) region locus was determined and 43 novel V H segments were identified. Instead, they refer to the sequence of the "coding" or "sense" strand: the one that's almost identical to mRNA-the difference of course being that every T in DNA is replaced by a U in RNA. But that's an inconvenient way to talk about a protein-coding DNA sequence: everything's not only complementary but also backwards.įor the sake of ease and clarity, scientists tend to ignore the bottom strand (they call it the "non-coding" or "antisense" strand). It would be more accurate to say that the DNA sequence of the "start codon" on the bottom strand is CAT. That means we'd have to write the sequence of the bottom strand like this: The scientific standard is to write a nucleotide sequence from 5' to 3'. That is, the 5' (5-prime) and 3' (3-prime) ends of the two DNA strands face in opposite directions: The chemical structure of DNA gives it a polarity, and the two complementary DNA strands are anti-parallel. While our shorthand version shows just the top strand, it's actually the bottom strand that RNA polymerase reads to build an mRNA molecule. If we wanted to, we could include the sequences of both strands: It's a shortcut, and it's tidier to look at, and it's how DNA sequences are typically written. It's not a mistake when we say that ATG is a start codon. The cell will keep reading until it reaches a stop codon, which may happen earlier or later than in the original sequence. A frameshift changes the grouping of bases into codons, affecting all the amino acids downstream of the mutation. Unless they happen in multiples of 3 bases, insertions and deletions shift the reading frame-which is why they’re also called frameshift mutations. Deletion mutations remove one or more DNA bases. Insertion mutations add one or more DNA bases. Silent mutations code for the same amino acid as before.Any codons after that are not translated, and the resulting protein is missing amino acids. Nonsense mutations make a premature "stop" codon.Missense mutations cause a single amino acid change in the protein.Changes to a gene’s DNA sequence, called mutations, can change the amino acid sequence of the protein it codes for-but they don’t always.Ī point mutation is a change to single DNA letter. Mutation is a process that causes a permanent change in a DNA sequence. If many applications are running at one time and you’re not using them all, consider quitting the unused apps. While fragmented HDDs are often the cause of performance slowdowns on Macs, there are other reasons a Mac might be sluggish. What Else Could Be Causing A Mac To Run Slowly? It also has other uses like creating a secondary drive so that the main drive can be defragged and optimized, finding and eliminating duplicate files, performing secure erasure of drives, and cloning drives. (Drive Genius 5 – note that the current boot drive can’t be defragged.)ĭrive Genius 5 is used by Apple employees at the Genius Bars and Genius Groves at Apple retail stores, so it’s highly regarded in the industry. Note that the utility does not work with APFS-formatted drives, but as noted in the previous section of this article, APFS includes an automatic defragging capability. The solution is to use Drive Genius 5 from Prosoft Engineering (see image below). Those creatives we talked about earlier can regain performance by defragging those large HDDs filled with big files, but Apple does not build a defragging utility into macOS. For HDDs - which as of publication are currently not supported with APFS - the new file system will provide automatic defragmentation in background when your Mac is idle. ![]() ![]() Never defrag an SSD!Īs of the release of macOS 10.13 High Sierra in September of 2017, some Macs now use a totally different file system called APFS (Apple File System) to replace the old HFS+ file system that had been in place for decades. Defragging an SSD or Fusion Drive (which is a combination of an SSD and a HDD) can actually be dangerous to the drive and reduce its lifespan. An SSD uses flash memory to store data rather than spinning mechanical platters. More and more Macs are doing away with Hard Disk Drives and using solid-state drives - SSDs - for data storage. HFC still does a very good job of keeping the files unfragmented, but as time passes this type of user may see a performance hit. Generally, these people are creatives who use a lot of big files in their work. The exception is for those Mac users who have hundreds or thousands of very large files (usually audio or video files) larger than 1GB in size. HFC works very well for most users and as a result, most users should never need to defrag a Mac hard drive. Think of HFC as constantly performing defragmentation on hard disk drives, ensuring that the HDD is always as optimized as possible for reading and writing data. Since the days of Mac OS X “Panther” (which shipped in 2003), Mac OS X and its successor macOS have used something called Hot File Adaptive Clustering (HFC) to automatically defragment files. Why Macs Usually Don’t Need to be Defragged ![]() The result? Faster loading of apps and data. This slows down operation.ĭefragmentation reads the files on the HDD platters, then rewrites them on the platters so single files are written contiguously and the drive head doesn’t need to move back and forth as much. As a result, those files are stored in locations all over the drive platters and the moving read/write head must travel the complete width of the platter to load files. ![]() Data can’t automatically migrate itself to the outer edges of the platters to fill in available space, so after many read/write cycles have been completed, there are gaps where large files cannot be written. New data is generally written to the outside of the platters, with the read/write heads moving in toward the center as more data is stored. HDDs use a magnetic read/write head to access rotating round platters that usually spin at either 5,400 or 7,200 revolutions per minute (see image above right). In this article, we’ll discuss why defragging Mac HDDs is usually unnecessary and how to do it if you’re one of the few users who may actually benefit from the process.ĭefragging is a slang term for “defragmentation”, which is the process of organizing files on a HDD to optimize how it accesses those files. One can maybe understand Chivalry having a certain goofy, Monty Python and the Holy Grail-esque charm as a free Half Life mod, where flat and ugly textures, horrible figure modeling, a choppy frame rate - especially bad on the Xbox One - and poor animations, clipping issues, and terrible hit detection could be laughed off because the conceit was pretty good. Unfortunately, that doesn’t seem to be the case, and Hardsuit Labs’ port of Chivalry: Medieval Warfare is a pretty disappointing mess on Xbox One, and only marginally better on the PS4. While the port to Xbox 360 and PS3 was not well received due to technical issues - from lag and framerate problems to sound bugs - there was hope that the game’s port to current gen consoles would address these faults and perhaps update it to recent standards. A multiplayer-only game, Chivalry transposes standard team shooter game types - deathmatch, team deathmatch, CTF and other multi-objective-based modes - to the Middle Ages. Focusing on melee combat and replacing modern weaponry with medieval armor, maces, swords, pikes and axes, the PC version has cultivated a large and devoted following, albeit one with a reputation for unpleasant online behavior like team killing.
